Specific antibody to neuropeptide

はじめに

　von EulerとGaddum^1）がウマの脳と消化管にsubstance Pの存在を示唆して以来，50年を経た今日，数々の生理活性ペプチドが脳と消化管に共存することが免疫化学的な手法により明らかにされてきた。もともと消化管組織から単離されたペプチドの免疫活性が中枢で検出され，一方，脳から単離されたペプチドの数々が胃・腸・膵で見出された。これら脳と腸管に共存するペプチド，すなわち脳—腸管ペプチドの発見は免疫組織化学あるいはラジオイムノアッセイ（RIA）などの免疫化学的研究における大きい成果の一つである。

　これら脳—腸管ペプチドが脳と消化管において，一致する化学構造を持つペプチドとして存在するか否かは興味ある問題である。すでにsubstance P，ニューロテンシン，ソマトスタチン，CCK-8およびVIPなどについては脳および消化管の両者から分離，精製され，抽出に用いた組織の相違にもかかわらず，一致したアミノ酸配列であった。他方，GRP，ガストリン，モチリンなど消化管由来のものは脳で免疫活性が検出されたにすぎず，終局的には免疫活性物質の単離，構造決定による同定が必要である。

　Region-specific antisera to neuropeptides have become important in biochemical and physiological studies on neuropepticles. Since an antiserum raised against a certain biologically active peptideconsists of, in most cases, heterogeneous antibodies recognizing different immunologic determinants of the peptide molecule, synthetic peptides related to the respective determinants in the peptide molecule are expected to be useful as artificial haptenic immunogen for production of the region-specific antisera.
