Alzheimer's disease (AD) is by far the most common cause of dementia in humans, and has been found to be etiologically heterogenous. Mutations in a single gene, such as the amyloid precursor protein gene (APP), presenilin-1 gene (PS-1) or presenilin-2 gene (PS-2), have been identified as the causative mutations of early-onset familial AD (EOFAD).

Sporadic AD and late-onset familial AD are the most common forms of AD, but their etiology is not clear.
