多因子疾患としてのアルツハイマー病

　アルツハイマー病の分子遺伝学的な研究は，当初単一遺伝子病としての見地から，主に早発型の家族性アルツハイマー病においてなされてきた。

　一方近年，本症の大多数を占める孤発例あるいは晩発型の家族性アルツハイマー病は，複数の遺伝学的因子や環境要因が関与して発症に至る多因子疾患であるとの見地からの研究が注目を集めている。アポリポ蛋白Eの遺伝子多型の一つであるAPOE4は，遺伝学的危険因子として代表的なものであるが，その他の危険因子についてもとくにAPOE4との関わりから現在精力的な研究が進められている。

Alzheimer's disease (AD) is by far the most common cause of dementia in humans, and has been found to be etiologically heterogenous. Mutations in a single gene, such as the amyloid precursor protein gene (APP), presenilin-1 gene (PS-1) or presenilin-2 gene (PS-2), have been identified as the causative mutations of early-onset familial AD (EOFAD).

Sporadic AD and late-onset familial AD are the most common forms of AD, but their etiology is not clear.
