GABA and epilepsy.

抑制性神経伝達物質GABAの機能不全は神経ネットワークの過剰興奮状態をもたらし，ひいてはてんかんにも関係すると考えられる。遺伝子ターゲッティングで作成されたマウスのうち，GABA合成酵素のGAD65，その補因子PLPの供給に必要なアルカリフォスファターゼ，GABA_Aレセプター・サブユニットのうちのβ₃のノックアウトでは，脳内GABA量またはGABAレセプター機能の減少とともに，てんかん様発作がみられた。またGABA系と神経機能とに異常がみられるミュータントのマウスと線虫についても説明した。

Any disorders of GABAergic functions in the central nervous system are implicated in epilepsy. Several lines of mice show spontaneous seizure when their genes encoding GABA-related proteins are inactivated by homologous recombination. In addition to seizure, the reduction of GABA contents is observed in mice lacking a 65 kDa isoform of glutamic acid decarboxylase and a tissue non-specific alkaline phosphatase, and the reduction of neuronal GABA sensitivity is accompanied with deletion of GABA_A receptor β₃ subunit. Impairment of motor function and GABAergic function is described in several mouse and nematode mutants.
