The formation and maintenance of myelin sheath in the central nervous system is the major function of oligodendrocyte. The death of oligodendrocytes due to the physical, chemical and biologi-cal factors including viral infection and immunological abberration plays a central role in the damage of myelin sheaths. There may be two possible mechanisms involved in the demyelinating processes associated with virus infection : a direct and an immune-mediated demyelination.

The first mechanism is an oligopathic virus infection in which the virus specifically attacks oligo-dendrocytes. The virus replicates in the oligodendrocytes, leading to the loss of ability to maintain myelin sheaths with consequent myelin destruction. Progressive multifocal leukoencephalopathy is a typical example of direct cytopathic diseases of the virus in which JC papovavirus attacks oligodendro-cytes without any immunological response.
