 With recent progresses in molecular genetics, a number of causative genes for hereditary diseases have been identified, and it is imperative to analyze individual genes from a large number of patients immediately and accurately. For this purpose, we are constructing high throughput gene analysis system for neurodegenerative diseases using GeneChipTM CustomSeq resequencing microarrays(Affymetrix), which can analyze 30 kilobases at one time with simple experiment protocols. We validated the accuracy of these microarrays to identify individual mutations of SOD1gene using genomic DNA samples obtained from patients with familial ALS. The results indicated that this microarray system was a robust tool for identifying point mutations, and potentially useful for deletion mutations. Improvement in analysis algorithms and application of robotics are future issues to be solved. The high throughput gene analysis system using DNA microarrays is thought to be useful not only for making quick and accurate diagnosis as well as promoting genomic medicine in clinical practice, but also for applying to explorative researches such as comprehensive analysis of disease-related genes for neurodegenerative disorders.
