　近年，遺伝性神経疾患の原因遺伝子が次々と同定されており，またポストゲノム時代の到来に伴うゲノム医学の発展とも相まって，臨床の現場でも簡便にかつ迅速に，遺伝子解析を行う必要性が高まってきている。そのため筆者らはDNAチップを用いて，神経変性疾患関連遺伝子の塩基配列を，網羅的にかつハイスループットに解析するシステムの構築を行っている。DNAチップは1度に3万塩基の配列が解析可能であり，複数の遺伝子を搭載して同時に解析することができる。チップの変異検出の正確性を，家族性ALSにおける既知のSOD1変異を用いて検証した。既知の変異はいずれも正確に検出可能であり，遺伝子変異/多型検出におけるチップの正確性が示された。DNAチップは，臨床において極めて有用なハイスループットな遺伝子変異解析システムを可能にするのみならず，疾患関連遺伝子の，体系的ゲノム解析といった研究面へも応用可能であると考えられる。

　With recent progresses in molecular genetics, a number of causative genes for hereditary diseases have been identified, and it is imperative to analyze individual genes from a large number of patients immediately and accurately. For this purpose, we are constructing high throughput gene analysis system for neurodegenerative diseases using GeneChip^TM CustomSeq resequencing microarrays（Affymetrix）, which can analyze 30 kilobases at one time with simple experiment protocols. We validated the accuracy of these microarrays to identify individual mutations of SOD1gene using genomic DNA samples obtained from patients with familial ALS. The results indicated that this microarray system was a robust tool for identifying point mutations, and potentially useful for deletion mutations. Improvement in analysis algorithms and application of robotics are future issues to be solved. The high throughput gene analysis system using DNA microarrays is thought to be useful not only for making quick and accurate diagnosis as well as promoting genomic medicine in clinical practice, but also for applying to explorative researches such as comprehensive analysis of disease-related genes for neurodegenerative disorders.
