Histological studies were made to locate indocyanine green (ICG) in pigmented rats using an infrared light microscope. In healthy rat eyes, ICG leaked from the choriocapillaris into the choroidal stroma. ICG did not leak through retinal vessels. It did not immigrate through the retinal pigment epithelium (RPE) anteriorly. In moderately photocoagulated fundus, the lesions showed hypofluorescence in early-phase ICG angiogram due to filling defect in occluded choriocapillaris. Two weeks later, the choriocapillar-is was restored and the RPE was repaired and became hyperplastic. ICG angiography showed blocked fluorescence in the center of hyperplastic RPE lesions. RPE cells, when selectively damaged by ornithine, were stained by ICG. ICG also stained the sensory retina by immigration through the damaged RPE. These findings would provide morphological basis in the interpretation of ICG angiographic features in clinical situations.
