PCR法を用いた涙液における帯状ヘルペスウイルスの証明

Detection of vericella-zoster virus DNA in the tear fluid by polymerase chain reaction

　帯状ヘルペス性角膜炎の2症例の涙液から，polymerase chain reaction （PCR）法を用いてより簡便に，帯状ヘルペスウイルス（varicella—zoster virus：VZV）の検出を試みた。

　方法は，2症例の涙液をシルマー試験紙2枚を重ねて採取し，Sepa-GeneにてDNAを抽出した。PCR後，PCR産物を電気泳動した結果，642 bpにバンドが検出され，VZVの存在が確認された。シルマー試験紙を用いた本法は，DNAの抽出，PCR，アガロース電気泳動の過程を委託にて行うため，極めて簡便であり臨床上有意義なものになり得る。

We attempted to detect varicella-zoster virus DNA in the tear fluid in 2 patients with varicella-zoster keratitis using polymerase chain reactionamplification. DNA was detected in the tear fluid obtained by double Schirmer test strips from both patients. This method is easy and the results idicate that the polymerase chain reaction may be useful in confirming the clinical diagnosis of varicella-zoster keratitis.
