 Postoperative infections are an important cause of failure of orthopedic treatments, in particular, of operations related to joint prostheses. The purpose of this study was to examine the mechanical strength andin vitrorelease of antibiotic-loaded calcium phosphate bone paste as a good defect filler and antibiotic carrier. Vancomycin hydrochloride, teicoplanin, gentamicin sulfate, amikacin sulfate, and flomoxef sodium were the antibiotics used. The pastes were divided into two groups according to differences in consistency. Calcium phosphate bone paste of low consistency containing 2.5%wt and 5%wt antibiotics showed favorable compression strength above 50 MPa at day 5. The release rate and released amount of antibiotics from paste were drug-, load-, and consistency-dependent. Good release of vancomycin, teicoplanin, gentamicin, amikacin was obtained, but release of flomoxef was poor. This system can be applied in a variety of surgical situations by changing the consistency of the paste and the type and amount of antibiotics.
