　ヒトの脳における，アミロイド前駆体蛋白質（APP）のmRNAの局在を明らかにすることを目的として，本研究を行った。APPのmRNAが神経細胞内に発現することは知られているが，神経細胞以外の脳細胞における局在に関しては研究者の間で議論のあるところである。今回の検討で，アイソフォームに関して非特異的なAPPに対するPC RDNAプローブにより，全例で神経細胞のみならずアストロサイトやアルツハイマー病の老人斑に関連した反応性ミクログリアも点状に染色され，そこにAPPのmRNAが局在することが明らかになった。APP695に対するcRNAプローブでは，神経細胞とグリアがよりびまん性に染色された。

This study aimed to provide clear data on the localization of the mRNA for the amyloid precursor protein (APP) in human brain. It is known to occur in neurons but the possible localization in other type of brain, cells is controversial among several authors. A cDNA probe constructed by polymerase chain reaction for APP, which was not specific for a particular isoform, gave punctate staining and showed a clear localization not only in neurons but in astrocytes in all brains,, as well as in the reactive microglia associated with senile plaques in Alz-heimer disease. A cRNA probe gave more diffuse staining of neurons and giia.
