抄録 腫瘍の悪性化は,その膜表面構造に重要な鍵があるとされてる。今回,脳腫瘍の成因を解明すべく,膜表面構造に関係する荷電変化に着目し,実験を行なつた。新しい細胞電気泳動装置として無担体連続電気泳動装置を用いた。対象はrat gliomaとfibroblastの継代細胞を用い,培養時期,剥離法,電気泳動の細胞への影響などの予備実験を行なつた.次いで人脳腫瘍細胞では,4例の各種悪性度のgliomaと,3例のmeningiomaを単層培養し,予備実験のデータに基づいて,これらの人脳腫瘍細胞の細胞電気泳動実験を行なつた。これらの結果は悪性細胞系ほど移動度が小さく,いままでに報告のある従来の細胞電気泳動実験結果と異なつた。しかし,我々の結果は悪性系細胞ほど陰性荷電が小さいことを示しており,この事は腫瘍の悪性化に伴なう膜の糖鎖不全現象という理論を裏づけていると思われた。また,これらの結果は臨床的にも,診断や治療方針の方向づけの一助となり,また,細胞の悪性化解明の一画をとらえているものと考えられた。今後の問題として,細胞膜荷電に関係する膜構造の検索が必要と思われた。
Malignant transformation in tumors is charac-terized by changes in cell surface components such as glycolipids and glycoproteins and is determined by the sialic acid content of the glycoconjugates. Our purpose was to investigate whether or not a similar change occurred in the brain tumor cells during malignant transformation. Change in the electrical charge of the cell surface was studied by means of cell electrophoresis.
Experimental cell lines of rat gliomas and fibro-blasts were cultured and compared for electrical charge by cell electrophoresis. Four various grade gliomas and three meningiomas from human sub-jects were cultured and compared for electrical charge by cell electrophoresis. Free flow electro-phoresis (Elphar Vap 5) was used because it chara-cteristically does not interfere with the use of the cells for biochemical and biological studies follow-ing electrophoresis and because it also allows thepossibility of subsequent cell culture.
The rat glioma cells had lower electrical charges than the fibroblasts, and the human malignant brain tumor cells exhibited lower electrical charges than the benign tumor cells.
These findings suggest a lower sialic acid content in malignant than in benign tumors and agreewith theories which postulate a decrease in sialic acid content accompanying malignant transforma-tion. This differentiation between benign and malignant tumors was made on the basis of the cell surface electrical charge and could be used for clinical diagnosis and treatment.
