Pyrithioxinの中枢作用機序に関する電気生理学的研究—ウサギの覚醒反応と誘発筋放電を対象として

ELECTROPHYSIOLOGICAL STUDIES OF THE EFFECTS OF PYRITHIOXIN ON THE CENTRAI NERVOUS SYSTEM

緒言

　Pyrithioxinは既に我が国においても脳卒中後遺症や頭部外傷後遺症などの治療に脳代謝改善剤として使用されている薬物である。そしてその作用機序についても代謝面についてはかなりの研究が行なわれているようであるが，脳代謝改善剤でありながら，脳の神経生理学的な面についての研究はあまり行なわれていないようである。本研究はこのようなPyrithioxinの神経系に対する作用機序を，特に意識と運動面について電気生理学的な面より検討した。

A study was made of the arousal reaction and evoked muscular discharge in rabbits in an effort to clarify the functional mechanism of Pyrithioxn on the central nervous system. The results obtained are as follows ;

1. The thresholds of arousal reaction in the cerebral cortex and hippocampus as well as that of evoked muscular discharge in the fore and hind limsb due to stimulation of the reticular formation were decreased following intravenous or oral admi-nistration of Pyrithioxin.

2. Pyrithioxin showed a facilitatory effect on the evoked muscular discharge in the fore and hind limbs due to stimulation of the ceredral cortex. It showed a facilitatory effect at a small dose and an inhibitory effect at a large dose on the discharge due to stimulation of the hippocampus.

Thus, it was strongly suggested that Pyrithioxin might facilitate consciousness and pyramidal move-ment.
