Antibody against the microsomal fraction of brain tumors was immunologically studied. Brain tumor homogenate in 0.25 M sucrose was centri-fuged 500g×5min, then the supernant was recen-trifugad 105,000g×60min. The pelet contained the crude microsomal fraction of the tumor was suspended in saline and injected intramascularly and in the foot pad of rabbits. The antiserum was absorbed by non-tumor brain tissue (tissue obtained from arterio-venous malformation) homogenate.

The antiserum was then separated by DEAE cellulose column chromatography. The immuno-globlin was tested with the tumor soluble protein, patient's cerebrospinal fluid and the serum. Using the Ouchterlony's method and immunoelectro-phoresis, the immunoglobulin reacted with cere-brospinal fluid, soluble tumor protein, but not with serum.

The titre of cerebrospinal fluid against the im-munoglobulin was tested by Ouchterlony's method. Astrocytoma, glioblasma patient's cere-brospinal fluid showed the titre of 16×to more than 512×against antiastrocytoma serum. Cere-brospinal fluid from head injury, cerebrovascular disease, pituitary adenoma, craniopharyngioma or pineal teratoma and meningioma showed the titre less than 8×. The titre has shown no correlation with cerebrospinal fluid protein content. Anti-meningioma antibody showed high titrea gainst the cerebrospinal fluid from meningioma patient. These results suggest a posibility using the antibody as an useful tool of immunological diagnosis for brain tumors.

Astrocytoma cell culture was carried out in the Eagle's medium with 20% fetal calf serum. No cytological changes were observed after adding 5% normal rabbit serum, anti-normal brain or anti-meningioma antiserum absorbed with normal brain homogenate in the medium. However, cytolysis was observed after adding 5% of anti-astrocytoma antiserum absorbed with normal brain homogenate. This observation suggest the specificity of the antibody.
