  • Look Inside

Effects of the central nervous system stimulants, including ANP-235, pipradrol, pheniprazine and ami-triptyline on the oxygen consumption and C14-leucine incorporation into protein of the brain cortex and liver slices of guinea-pig were studied. Effects of these drugs on the oxidative phosphorylation were also ex-amined using mitochondria isolated from brain cortex.

1) ANP-235 inhibited the labelled amino acid in-corporation into cerebral protein at lower concentration, where no inhibitory effect was observed on the oxi-dative phosphorylation.

2) Pipradrol showed stimulatory effect on the ami-no acid incorporation, but no effects were obtained on the oxidative phosphorylation.

3) Pheniprazine inhibited both the respiration and the amino acid incorporation at the same degree in proportion to the drug concentration.

4) Amitriptyline inhibited markedly the amino acid incorporation at the concentration where no respiratoy inhibition were observed. Its uncoupling effect on the oxidative phosphorylation may be responsible for this phenomenon in the protein synthesis.

5) Biochemical mechanism of the effects of these drugs on the central nervous system were discussed.
