By the use of agar-gel-diffusion-precipitin method, specific antigens were immunochemically sought in human gliomas and rat brains.

Antigens were extracted from the tissues by pH 4.8 acetate buffer according to the method originated by H. Hirai et al. and were injected to rabbits with Freund's complete adjuvant. Immune-sera were obtained from the rabbits two to three months later and applied to the agar-gel-diffusion-precipitin test.

Of three cases of human gliomas investigated, a specific antigen was demonstrated :n a case of glio-blastoma mutiforme, the most malignant type of gliomas, as a clearly visible precipitin-line on agar plates.

The line was shown to appear exclusively against the antigen of this tumor and not against the antigens of the normal human brains, liver and kidney as well as of other gliomas. This tumor-specific precipitin-line was not stained by fat stain and Schiff's stain. From the absorption-spectrum of utraviolet rays, this antigen seemed to be a simple protein in nature. In the other two gliomas of comparatively benign type, tumor-specific antigens failed to be demonstrated.

An organ-specific antigenic protein of the rat brain was demonstrated by the same method and its natures were investigated. The protein was very soluble in water, immigrated in the electric field with the same mobility as that of rat serum alpha-globulin at pH 8, and precipitated by ammonium sulfate between the concentration of 23% and of 32%. This specific pre-cipitin-line was not stained by fat stain and Schiff's reagent. By subcellular fractionation the specific antigen was proved to exist in the nuclear fraction.
