猫に皮質皮質下慢性電極を植込み,imiprami-neの脳波と行動に及ぼす影響を観察した。imi-pramineは行動面では鎮静作用を示す。脳波は全体に徐波化する。脳幹網様賦活系の興奮性は低下し,辺縁系の興奮性は上昇する。かかる成績は近年明らかにされた脳幹網様系並に辺縁系の生理学的知見と併せて考えるならば,imipramineがうつ状態の症状改善に有効に作用する薬理学的基礎を提出するものと考えられる。
In 1957, R. Kuhn reported that imipramine (tofranil) (N-(γ-dimethyl-amino-propyl) imino dibenzyl hydrochloride) had good effects on many cases of depressive states. Since then many authors, including one of the authors of this report, Z. Kaneko, have reported its clinical value for depressive states. To find the mechanism of its action, its effects on EEG and the behavior of the cat were stu-died.
Cortical and subcortical electrodes were implanted chronically with stereotaxic appa-ratus of the Noken type and by the aid of the map of Jasper and Ajmone-Marsan.
More than one week after the operation, 8-25 mg/kg imipramine (mainly 15 mg/kg.) was injected into the peritoneal cavity and its effects upon the following points were observed: (1) behavior (2) spontaneous EEG activity (3) cortical desynchronizing respon-se to electrical high frequency stimulation (Imsec square wave, 100 c/s, 5 sec) of the brain stem reticular formation (4) recruiting response to low frequency stimulation (Im-sec 6-10 c/s) of the thalamic diffuse activa-ting system, (5) seizure after discharge fol-lowing high frequency stimulation (Imsec 100 c/s 5 sec) of the limbic system (hippoca-mpus and amygdaloid nucleus)
