External signals such as hormones, neurotransmitters and other molecules are transmitted into cells through several signal transduction pathways, resulting in the production of second messengers, to exert their biological actions. Many second messengers are poduced from membrane phospholipids by the action of phospholipases, effector enzymes. The well-known phospholipase as an effector enzyme is phosphoinositide-specific phospholipase C which produces two second messengers, inositol 1,4,5-tris-phosphate and diacylglycerol. Phospholipase A2 also seems to be an effector enzyme, since arachidonic acid produced by this enzyme from arachidonate-containing phospholipids acts as a putative second messenger. In addition, it has been found that phospholipae D couples with agonist receptors to produce phosphatidic acid which has second messegner-like activity. In this review, characteristics of these phospholipases and their activation mechanisms in response to agonists are described.
