A MODIFIED IMPREGNATION METHOD FOR STAINING APICAL DENDRITE. AND SPINE (GOLGI MODIFICATION METHOD)

I．はじめに

　Apical dendriteおよびspineの同時検出のためのGolgi法の変法を報告する。Golgi法^2）では新鮮材料を用いる必要があるが，木変法では長期間Formalin内に貯蔵されたautopsy materialに於ても，比較的安定した結果がえられる点，有効な染色法と考えられる。

A silver impregnation method for the demon-stration of apical dendrites and spines in materialslong preserved in folmalin is reported. This methodis a modified version of Golgi's Silver method andconsists of the following procedures:

in a solution containing 3% potassium bichromate(150ml), 96% ethyl alcohol (50ml), and concentratedacetic acid (50ml) for 24 hrs. at 37℃, and 4 moredays at room temperature after changing the sol-ution twice. The tissue blocks were then keptfor 5 days at room temperature in a 5 % potassi-um bichromate solution. After making a thoroughrinsing for one day in tap water and three washi-ngs in distilled water, the blocks were impregna-ted for 1/2 to 1 hr. in a 0.5% AgNO₃ solution,for a further 4 days at room temperature in a 1 %AgNO₃ solution, washed three times briefly indistilled water and again impregnated for 4 to 5days in a 2% AgNO₃ solution. Following a tho-rough rinsing in distilled water the tissue blockswere embedded in paraffin and 50 to 100μm sectionswere prepared.
