It is said that nitrazepam induces sleep by means of exerting inhibitory influence on limbic system and hypothalamus which are closely related with emotion, and can keep the rhythm of wakefulness and sleep normally, while other commonly used hypnotics give inhibitory effects on reticular for-mation and neocortices.

We made experiments with cats for ascertaining the nature of action of nitrazepam and observed clinical effects on subjects who complained of sleep disturbances.

The results obtained are as follows.

1. Nitrazepam exerted the influence on the rhythm of wakefulness and sleep of free cats, name-ly, depressed appearance of wakefulness period and paradoxical sleep period.

2. After injection of nitrazepam, about 15 c/sec fast waves became dominant in drowsy period, and in slow wave sleep period, the same waves were al-so observed at intervals when the spindle waves did not appear. Hippocampal rhythmic waves which appeared in the period of paradoxical sleep dimini-shed its frequency to 3~3.5 c/sec, making a 'striking contrast to the frequency usually shown in control study (6~7 c/sec).

3. Nitrazepam diminishes the amplitude of nega-tive waves of recruiting responses which are evoked on cortices by stimulation of nonspecific thalamic nuclei. This drug, therefore, is considered to exert influence on the diffuse thalamo-cortical projection system.

4. From viewpoint of the clinical study, this drug is confirmed to be suitable for insomnia based on neurosis and slightly depressive state. But, be-cause of its weak actions it may be seldom effect-ive on pathologic insomnia, such as caused by de-pressive state, manic state and schizophrenia.
