 N-propyl-N'-nitro-N-nitrosoguanidine (PNNG), a weaker gastric carcinogen than MNNG, was administered to Wistar male rats at a concentration of 100 μg/ml in the drinking water. Microcarcinoma in rats were defined as “intramucosal carcinomas with a diameter of less than lmm”. In this study, erosion and regeneration of the gastric mucosa were rarely found during PNNG administration, but microcarcinomas and invasive carcinomas were found in the glandular stomach of rats in about week 70. Four intramucosal carcinomas were found in weeks 67~88, and three of them were microcarcinomas. Microcarcinomas of 0.5~0.6 mmin diameter showing glands with cellular and structural atypism had spread by infiltration in the mucosa. These lesions were surrounded by normal pyloric mucosa. Differentiated adenocarcinomas invading the submucosa were found in weeks 69 and 70, and signet-ring cell carcinomas invading the serosa were found in week 88.

 In humans, microcarcinomas are frequently surrounded by intestinalized epithelium. In this study, intestinal metaplasia was first found in week 19 and its appearance preceded that of microcarcinomas. All rats with microcarcinomas also had intestinal metaplasia, but these two types of lesions were in separate areas of the stomach.
