 To investigate genetic characteristics of multiple colorectal carcinomas (CRC), K-ras mutation, LOH (loss of heterozygosity) of chromosome 17p, LOH of chromosome 18q, MSI (microsatellite instability) were studied by molecular biological methods. K-ras codon 12 mutation was analyzed by Mutant-Allele-Specific Amplification. The frequency of K-ras mutation in multiple CRC was 12.0%, which is lower than that in single CRC (40~70%). The frequency of MSI in metachronous multiple colorectal carcinomas is 25.0, which is higher than that in synchronous multiple CRC (4.2%) and single CRC (6.7%). These data suggest that multiple CRC may develop a different genetic pathway with lower K-rus mutation and that metachronous CRC may include HNPCC (hereditary nonpolyposis colorectal cancer).
