大脳皮質培養神経細胞を用いてのけいれん発現機序の解明

Studies on the mechanism of seizure discharge using cerebral cortical cultured neurons.

I．はじめに

　けいれん波が大脳皮質に起こっているときには，神経細胞にはbusting activityとかparoxysmal depolarization shiftといわれる特徴的な神経細胞膜電位変化が必ずみられる。われわれはこれまで，このbursting activityが起こっているときの神経細胞内でのカルシウム関連の一連の変化を，大型の神経細胞をもつミスジマイマイの神経節細胞の中で，けいれん剤ペンチレンテトラゾール（PTZ）にきわめて感受性の高い神経細胞を用いて検討してきた_6〜10,15）。

　今までにミスジマイマイのPTZ-感受性細胞とPTZ-非感受性細胞との比較をしながら，各種の実験を行なって得られた結果のあらましは次の通りである。

Our previous studies using snail neurons demonstrated the following results during bursting activity induced by pentylenetetrazol (PTZ) in PTZ-sensitive neurons of the Japanese land snail, Euhadra peliomphala. 1) During bursting activity, intracellular calcium moved toward the cell membrane areaeven after blockade of calcium influx. 2) In the Euhadra neurons, the source of calcium release was lysosome-like granules (LLG) and stored, calcium released with morphological changes of LLG during bursting activity.
