I．はじめに

　アレルギー性脳炎モデルとして，ここでは実験的アレルギー性脳脊髄炎（experimental allergic encephalomyelitis，EAE）と実験的アレルギー性神経炎（experimental allergic neuritis，EAN）を取り上げる。EAEは最初狂犬病ワクチンや日本脳炎ワクチンなど，脳を基剤とするワクチンを接種した場合に，副次的に起こるワクチン接種後脳炎（いわゆる後麻痺）の原因究明を目指して出発したモデルである。初期には，サルに脳乳剤そのものを頻回（たとえば週3回ずつ40回以上）皮内接種し，100日以上の日数をかけて誘起実験を行なっていた^1,2）。が，1942年にFreund^3），Kabat^4），Morgan^5）が独立にEAE誘起にいわゆるフロイントの完全アジュバント法を導入するに及んで，少量の脳乳剤を用いて2〜3週間の短期間で発症させることができるようになって，研究は飛躍的に進歩した。1960年から1970年代にかけて抗原研究が神経化学的分析手法の発展とともに急速に進み，EAE抗原が中枢神経のミエリンに局在する塩基性蛋白質であることが判明，ついで，1971年に至って，Eylar^6）とCarnegie^7）によって独立にこの特異な蛋白質の全アミノ酸配列が解明された。

　The usefulness and limitations of experimental allergic encephalomyelitis (EAE) and experimental allergic neuritis (EAN) as animal models for the human demyelinating diseases are discussed with regards to multiple sclerosis (MS) and Landry-Guillain-Barre syndrome, respectively.

　Extensive immunochemical analysis has been performed on myelin basic protein (MBP) as EAE antigen. Thus, complete amino acid sequence, EAE-inducing sites, delayed type hypersensitivity-related sites and antibody production-related sites have been identified on MBP molecules by several workers.
