In cell free materials of calf splenic nerve obtained by Euler's press juice method, in isotonic potassium phosphate (pH 7.0), a sediment rich in noradrenaline could be obtained by high speed entrifugation. An active fraction rich in SP was also obtained by high speed centrifugation of press juice from bovine vagus nerve or spinal dorsal root.

Such particulate SP and/or noradrenaline was released not so easily by treating with isotonic medium at neutral pH in the cold. But, it could he liberated with relative repidity by raising tem-perature, acidification and hypotonicity.

Electron-microscopical observations were also made on these nerve axons and their subcellular fractions. The axon has shown by a longitudinal section the well known structure with elongated mitochondria and uncounted neurofilaments. It was confirmed that the vesicular structure with a mean diameter of 1,000 Å were also seen in the axon of vagus nerve and spinal dorsal root, and moreover, in the fraction of high speed sediment.

It is concluded that noradrenaline and SP which have been considered as transmitter substance were Present in high concentration in particulate structures in the peripheral nerve.
