培養骨格筋における電気興奮性の発達

Development of excitability in cultured skeletal muscle cells

はじめに

　骨格筋細胞は収縮機能を果たすべく高度に分化した細胞である。その細胞膜には大きな細胞内陰性の静止電位が存在し，また静止電位を少なくするような各種刺激に応じてその膜電位が一過性に細胞内陽性に逆転する，すなわち活動電位を発生する性質をもっている。脊椎動物骨格筋における収縮は通常の場合この活動電位によって誘発されている。このような顕著な膜の性質が筋細胞の分化発生の過程においてどのように獲得されてくるかについては近年に至るまであまり知られていなかった。これを追跡することは，成熟筋細胞膜における電気興奮性の機構を明らかにするためにも，また細胞分化の本態を探るためにも非常に有効なことであると思われる。

　静止電位や活動電位はHodgkin-Huxley説によれば膜を隔ててのイオンの濃度勾配と膜の選択的イオン透過性に依存している。すなわち静止時においてはKイオン透過性が優性で，膜電位は細胞内負のKイオン平衡電位に近く，刺激によって脱分極されると，Naイオン透過性が急激に増大し，膜電位はNaイオン平衡電位に近づき，活動電位を発生する（Hodgkin，1951）。

The surface membranes of mature skeletal muscle cells are specialized to maintain relatively large transmembrane electrical potentials and to generate action potentials. The magnitude of both the resting and action potentials depend upon ion-selective channels in the membrane, and upon ion gradients across the membrane. While these properties have been widely studied in the adult skeletal muscle, little was known regarding the development of these properties during muscle cell differentiation.
