Sarcoplasmic reticulum (SR) has two main functions in excitation-contraction coupling of skeletal muscle. One is to release calcium ion into sarcoplasm to initiate contraction. Another is to uptake calcium into SR space for relaxation of muscle. The mechanism of calcium uptake has been analyzed mainly on the fragmented SR preparations, which are isolated by the differential centrifugation. However, caffeine, which is known to release calcium from SR, is less effective in fragmented SR than in intact muscle.
