Fabry病の遺伝生化学的研究

Biochemical and genetic studies in Fabry's disease

I．まえがき

　近年，生化学の進歩により種々の遺伝性代謝異常疾患の病因が明らかになってきた。

　遺伝性代謝異常疾患の一つであるFabry病は1898年にイギリスのAnderson^1）とドイツのFabry^2）によってほとんど同時に報告された。Anderson^1）は被角血管腫，蛋白尿，手指変形，浮腫などのある症例をangiokeratomaの1例として報告し，Fabryは皮膚血管腫，蛋白尿などのある患者を初めpurpura hemorrhagica nodularisとして報告し，後にangiokeratoma corporisdiffusumという疾患名を提唱した。その後，1909年にはSteinerら^3）が本症に無汗症，間歇性四肢末端部疼痛があることを報告し，1925年にWeicksel^4）が本症に角膜混濁と結膜および網膜血管異常のあることを報告した。1947年Ruiterら^5）が本症の剖検を行ない，storage diseaseであると報告した。その後外国では多くの報告がある^6）。

Genetic, biochemical and enzymatic studies were performed in two families with Fabry's disease in order to obtain information on the relationship between the clinical manifestations and biochemical abnormalities. The leucocyte a-galactosidase, β-galactosidase, N-acetyl-β-glucosaminidase and N-acetyl-β-galactosaminidase activities were determined in 23 healthy adult controls, 10 members of family F and 17 members of family H. The urinary glycolipids were also measured in 10 members of family F and 17 members of family H.
