はじめに

　パーキンソン病は高齢者に頻度の高い神経変性疾患であり，全人口では千人に一人の有病率であるが，65歳以上の人口では1％以上が罹患するといわれる。臨床的には振戦，無動，固縮，姿勢反射障害などの運動障害を主とする神経症状が十数年にわたって進行し，末期には寝たきりになるという経過をたどる。神経病理学的には，中脳黒質のドーパミン作動性神経の選択的変性脱落が特徴である。

Autosomal recessive juvenile parkinsonism (AR-JP) is caused by mutations in the parkin gene. Parkin protein is characterized by a ubiquitin-like domain at its NH2-terminus and two RING finger motifs and an IBR (in between RING fingers) at its COOH terminus (RING-IBR-RING). We showed that Parkin is an E3 ubiquitin ligase which binds to ubiquitin-conjugating enzymes (E2s) through its RING-IBR-RING motif. The function of E3 is to recognize its specific substrate protein and promote its ubiquitination and subsequent degradation.
