神経移植によるパーキンソン病の治療―遺伝子治療に関連して

Neural transplantation for Parkinson's disease in relation to gene therapy

パーキンソン病に対する外科的治療法の一つとして神経移植はもっとも新しい分野であり，21世紀の再生医学の一翼を担うものとして期待されている。その主な目的は，欠落したドパミンを産生する細胞を脳内移植し宿主に供給することと，神経栄養因子を産生する細胞を脳内移植し宿主の内因性ドパミン神経系を活性化することにある。近年の分子生物学的手法の発達により，これらの目的を果たすための種々の細胞を作製することができるようになり，この分野も遺伝子治療としてのアプローチが重要になってきた。本稿では，われわれの研究を交えながら，これまでのパーキンソン病に対する神経移植の流れを整理し，今後の展望について遺伝子治療との関連にふれながら考察する。

Neural transplantation is one of the most promising approaches for the treatment of Parkinson's disease. This type of study started in the latter half of 1970s. Initially, dopamine neurons from the animal fetuses were used as donors and then paraneurons such as chromaffin cells were used. Based upon a large number of experimental animal studies, neural transplantation has been applied clinically.
