Parkinson's disease (PD) is characterized by cell loss in the substantia nigra and a reduction of dopamine in the striatum. Since dopamine does not cross the blood-brain barrier, it cannot be administered directly, but its precursor, levodopa (L-DOPA) can cross it. These led to clinical trials with levodopa (L-DOPA) in 1960s. Levodopa is an effective treatment of PD, but the side effects nausea, vomiting and involuntary movements are frequent and severe.
