Tyrosine kinase receptors uniquely expressed on motoneurons

はじめに

　運動ニューロン疾患（motor neuron disease：MND）は，運動ニューロンが選択的に変性をおこす疾患で，錐体路（一次ニューロン）および末梢運動神経（二次ニューロン）共に変性する筋萎縮性側索硬化症（amyotrophic lateral sclerosis：ALS），比較的良性の経過を辿るWerdnig-Hoffmann病やKugelberg-Welander病などを含む疾患単位である。本症の成因として，免疫異常^1-3），興奮性アミノ酸の代謝異常^4-7），金属や有機燐製剤などの神経毒性物質の関与，異常ニューロフィラメントの蓄積^8），家族性ALSにおける遺伝子異常^9-11）などが考えられているが，いまだ定説はなく，有効な治療法がみつかっていない。しかし近年，神経細胞の生存維持に関わる因子である神経栄養因子がいくつか同定されてきており，しかもMNDの動物モデルへの試験投与の有効性が認められるに及んで，神経栄養因子がMNDにおける治療方法として注目されるようになってきた。

Neurotrophic factors are promising drugs for saving motoneurons in motoneuron diseases. Motoneurons did not respond to nerve growth factor (NGF) and many attempts have been made to identify neurotrophic molecules for motoneurons. Ciliary neurotrophic factor (CNTF), brain-derived neurotrophic factor (BDNF), neurotrophin-3, -4 and-5 among the neurotrophins, insulin-like growth factor-1 (IGF-1), fibroblast growth factor 5 (FGF-5) and glial cell line-derived neurotrophic factor (GDNF) have been shown their trophic activities for motoneurons.
