5）ミトコンドリアDNA情報の発現調節

Regulation of the mitochondrial genetic expression.

はじめに

　ミトコンドリアは独自のゲノムをもち，核ゲノムとは半独立にDNAの複製，RNAとタンパク質合成を営んでいる。これらのミトコンドリアDNA（mtDNA）情報発現にかかわるタンパク質因子はすべて核ゲノムにコードされ，細胞質で合成され後にミトコンドリア内に移行する。そのため，mtDNA情報の発現調節には核遺伝子の関与が必須である^1）。

　また，mtDNA情報発現系は原核生物や真核生物の核遺伝子のそれと基本的に異なる点が多い。最近のミトコンドリア脳筋症の研究の進展に伴って，CPEOはmtDNAの大欠失^2），MELASとMERRFはそれぞれtRNA-Leu（UUR）とtRNA-Lysの点変異によって発症することが判明した^3～5）。さらに，それらはmtDNAの情報発現系の特殊性と密接に関連していることが明らかになりつつある。

Mitochondrion has its own genetic system but all the proteineous components are encoded by nuclear genes. Therefore, the mitochondrial genetic expression is regulated by the nucleocytoplasmic systems as well as the mitochondrial one. In addition, the mitochondrial genetic mechanisms are quite specific in some points as compared with the nuclear genetic one. First of all, mitochondrial DNA (mtDNA) is maternally inherited. Point mutations responsible to MELAS or MERRF were inherited from mother to children. In the most cases, the ratio of the mutant mtDNA/wild type of the children was more than those of the mothers.
