What is the molecular mechanism that determines the synaptic specificity? In this article, we review the findings obtained from a model system, the neuromuscular connectivity in Drosophila. We describe the identification and functional analysis of the target recognition molecules that are expressed in subsets of muscles and determine the neural specificity. We will also introduce recent finding that postsynaptic muscles extend dynamic protrusions during the period of neuromuscular recognition, and discuss its implication on the mechanism of synaptic specificity.
