We evaluated the effect of 5-fluorouracil (5-FU) as applied subconjunctivally following the first or second trabeculectomy in 41 glaucomatous eyes in 33 patients. The intraocular pressure (I0P) level equal to or less than 20 mmHg was adopted as the criterion for successful IOP control. The results were evaluated through life-table analysis and were compared with findings in 36 glaucomatous eyes that underwent first or second trabeculectomy without 5-FU treatment.

At 11 months after surgery, the probability of successful IOP control in 5-FU treated group was62% without medication, 83% with topical medica-tion and 91% with additional oral carbonic anhy-drase inhibitor. In the control group, it was esti-mated to be 19%, 63% and 77%, respectively. At the latest examination, the IOP of successfully controlled eyes averaged 9.9 mmHg in the 5-FU treated group and 14.2 mmHg in the control group. The difference was highly significant (p<0.001). Postoperative subconjunctival 5-FU administra-tion appeared to improve the prognosis following trabeculectomy performed as the first or the second procedure.
