複数のautocrine loop遮断によるヒト悪性glioma細胞の増殖制御効果について

Growth Control of a Human Glioma Cell Line by Multiple Autocrine Loop Blockade

　今回我々は，ヒト悪性glioma培養細胞において，抗成長因子特異抗体による複数のautocrine loopを遮断することにより，その増殖がどの程度抑制できるのかを検討した。まずcell ELISA法によりEGFR（epidermal growth factor receptor），FGF（fibroblast growth factor），PDGF（platelet derived growthfactor）をすべて分泌しているU251MG細胞を同定し，これに対してMTT assayおよび³H thymidineの取り込み試験を行った。その結果，上記の3種類の抗成長因子モノクローナル抗体をそれぞれ単独で作用させた場合，最大成長抑制効果は54.8〜69.0％（平均63.4％）であったが，3種類の抗体を同時に作用させた場合には，最大抑制効果86.3〜88.1％（平均87.2％）を得た。これらの結果より，抗成長因子特異抗体による複数のautocrine loopの遮断が，悪性gliomaの成長をより効率的に抑制し得ることが明らかにされた。

Autocrine growth due to dysregulation of growth factor production may have a role in the develop-ment of neoplasia. We demonstrated that U251MG, a well characterized human malignant glioma cell line, had high affinity receptors for epidermal growth factor (EGF), fibroblast growth factor (FGF), and platelet-derived growth factor (PDGF) by enzyme-linked immunosorbent assay. We asses-sed the inhibitory effect of and-EGF receptor (EGFR), anti-FGF, and anti-PDGF monoclonal antibodies (MoAbs) on the growth of U251MG cells using the MTT assay and ³H thymidine uptake.
