抄録 S-adenosyl-L-methionine sulfate tosylate (FO−1561)の脳虚血後の血流再開時における脳機能,脳エネルギー代謝に対する効果について検討した。ラットを用いて,four-vessel occlusionに低血圧を加えることにより,両側大脳半球の虚血を作製し,両側総頸動脈のクリップを解除し,血圧を虚血前の値に戻すことにより血流再開を行なった。脳波のpower spectrum分析を行なうと,血流再開後,FO−1561投与群では対照液投与群に較べて,徐波成分が少なく,速波成分が多い傾向がみられ,血流再開90分後のATP値が対照液投与群に較べて有意に高値を示した。
Effects of S-adenosyl-L-methionine sulfate tosy-late (FO-1561) on postischemic cerebral functionaland metabolic recovery in experimental cerebral ischemia were investigated.
Severe bilateral forebrain ischemia in rats was induced by four-vessel occlusion with reducing the mean arterial pressure to 100-110 mmHg. After forebrain ischemia had been maintained for 30 minutes, recirculation was started by removal of the arterial clamps of bilateral common carotid arteries and by increasing systemic arterial pressure to the preischemic level. The EEG was conti-nuously recorded from gold-coated screws inserted bilaterally in the parietal bones with the tips in extradural position, against a reference inserted prefrontal bone. Analysis of power spectrum of EEG activity was done by Berg Fourier Analyser. The brains were frozen in situ with liquid nitrogen before, during and after ischemia and then chisel-led out during irrigation with liquid nitrogen. Concentrations of ATP in brain tissue were de-termined with high performance liquid chromato-graphy. FO-1561, 100 mg/kg, was given intra-venously, immediately after recirculation.
After recirculation there was a tendency that EEG power spectrum in FO-1561-treated animals contained higher percentage of beta wave com-pared to that in control animals, while delta wave was lesser in FO-1561-treated animals. At 90 minutes following recirculation, ATP level in control animals was 2. 17±0.05 μmol/g (mean±SE) and 2.42±0.03 μmol/g (mean±SE) in FO-1561-treated animals. Thus, recovery of ATP level was significantly better in FO-1561-treated animals than in control animals (p<0.01).
