On the Cytochrome Oxidasc Inhibition of Chlorpromazine

　まえがき

　Phenothiazine系Alkylamine誘導体は主として抗ヒスタミン薬として研究されてきたが，Phenothiazine核にChlorを導入したChlorpromazineが精神神経疾患に効果^1）のあることが認められて以来，向精神（Psychotropic）薬として脚光を浴び，更にReserpine LSD-25，Serotonine等とともに，薬理学はもとよりその関連領域における基礎的研究とあいまつて臨床的にも精神病治療，病因解析に向つての有力な手段となつた。

　さて，Chlorpromazineの薬理作用については数多くの研究がなされており，その作用は極めて多岐に亙つている。例えばCouvosier^2）等は抗アドレナリン，神経節遮断，体温下降，抗ショック，抗痙攣，制吐，鎮痛，中枢抑制等の作用のあることを報告している。

　To take a step to the elucidation of the mode of action of chlorpromazine, its effects on cell metabolism with reference to those on substrate oxidation, dehydrogenase, oxidative phosphorylation and cytochrome oxidase were investigated from a biochemical standpoint. In an attempt to follow the mechanism of its cytochrome oxidase inhibition, it was suggested that chlorpromazine may attack Fe⁺⁺⁺ in the cytochrome oxidase reaction.

　These points in comparison with findings of other investigators will be discussed in this paper.
