Glial apoptosis and its importance in neurodegenerative disease

はじめに

　神経活動の「主役」が神経細胞であるなら，astrocyte，oligodendrocyte（oligo），microgliaの三種類の細胞群に大別されるグリア細胞は，神経活動の「裏方」を務める細胞である。「裏方」ではあるが，グリア細胞に異常をきたしたり，または欠落を生じたりすると神経機能は著しく障害され，神経疾患を生じることからもわかるように，その役割は重要である。こうしたグリア細胞の役割，とくにその生理学的，分子生物学的な機構について解らない点が多かった。しかし，最近の研究，とりわけ分子生物学，遺伝子解析法を用いた研究によって，今日，われわれの知見はかなり深まったといえる。たとえばastrocyteからは種々の神経栄養因子が放出されていること，およびそれらの作用機序も解った。また，oligoや，Schwann細胞は髄鞘を作り神経伝導を速める他に，栄養因子や毛様体神経節栄養因子（ciliary neurotrophic factor：CNTF）などを放出して神経細胞死を防いだり，あるいは神経突起の発芽・再生を抑制している役割があることなども明らかとなった。さらに脳内の掃除屋と考えられていたmicrogliaにも，栄養因子を出したりする一面や，一方神経細胞死を誘発することもあるなどの事実がわかりつつある。

Neurons and glial cells are the main components of the central nervous system. Glial cells play important roles in maintaining the functions of the central nervous system. For example, astrocytes produce various growth factors which maintain neuronal functions ; oligodendrocytes form myelin, which are important in impulse conduction and they inhibit the growth of neurons and astrocytes. Other glial cells include microglias, which function as phagocytes as well as to produce various trophic factors which are used in tissue repair.
