軟骨発生と破壊の分子機序

Bone and Cartilage Diseases and Regeneration. Transcriptional regulation of chondrogenesis and articular cartilage destruction.

　軟骨研究は，20世紀の培養細胞を用いた分化研究から，20世紀末から21世紀に入りマウスジェネティクスの手法を用いた生体での分子機序解明へと進んだ。軟骨組織発生のマスター遺伝子である転写因子が同定され，現在はそれら遺伝子を中心とした転写因子ネットワークの解析が行われている。さらに，生後の関節軟骨に発症する退行性変性疾患である変形性関節症の病態発症・進行の分子機序も次第に明らかになってきている。 　本総説では，最近の知見を交えながら，軟骨における転写因子群の役割を概説したい。

　In 20th century, cartilage researches were performed by in vitro culture system, and current trend of mouse genetic approaches makes a good progress in the studies of molecular mechanisms underlying chondrogenesis. Sox family and Runt family transcription factors play central roles in chondrogenesis and pathogenesis of osteoarthritis. In this review, we would like to introduce recent progress in understanding of molecular regulation of transcription factors in chondrogenesis and articular cartilage destruction.
