The transcriptome analysis revealed that >90% of human genomic regions produce functionally unannotated noncoding (nc)RNAs. In recent years, functional explorations have been unveiling the roles of these ncRNAs. One series of ncRNAs were shown to act as the site-specifying factor for epigenetic regulation with histone modification enzymes. Indeed, one of these ncRNAs is upregulated in the cancer cells in which it globally modulates the patterns of histone modification that may link it to cancer metastasis. Additionally, the association of the RNA-binding protein that causes a neurodegenerative disease with a specific ncRNA is markedly enhanced in the patient brain. In this chapter, the recent progress in the research of ncRNAs and their association with human diseases is introduced.
