Mechanism of the formation of β2-microglobulin amyloid fibrils

透析アミロイドーシスは，10年以上にわたって血液透析を受ける患者に共通してみられる深刻な合併症であり，アミロイド線維沈着の主な構成成分はβ₂-ミクログロブリン(β₂-m)である．生体内におけるβ₂-mアミロイド線維の形成機序を解明することは，透析アミロイドーシスの予防法や治療法の開発に重要である．β₂-mアミロイド線維の沈着は骨関節組織に多くみられることから，グリコサミノグリカンやプロテオグリカンのような骨関節組織の細胞外マトリックス分子などが，アミロイド線維の形成と沈着にかかわると考えられる．

β₂-Microglobulin (β₂-m) has been identified as a major structural component of amyloid fibrils deposited in dialysis-related amyloidosis, a common and serious complication among patients receiving hemodialysis for more than 10 years.　Elucidating the mechanism of the formation of β₂-m amyloid fibrils under physiological conditions is essential for developing the prophylaxis and therapeutics of dialysis-related amyloidosis.　Because the deposition of β₂-m amyloid fibrils is observed predominantly in the osteoarticular tissues, the extracellular-matrix molecules in the tissues, such as glycosaminoglycans and proteoglycans, may affect the formation and deposition of fibrils.
