β2-Microglobulin (β2-m) has been identified as a major structural component of amyloid fibrils deposited in dialysis-related amyloidosis, a common and serious complication among patients receiving hemodialysis for more than 10 years. Elucidating the mechanism of the formation of β2-m amyloid fibrils under physiological conditions is essential for developing the prophylaxis and therapeutics of dialysis-related amyloidosis. Because the deposition of β2-m amyloid fibrils is observed predominantly in the osteoarticular tissues, the extracellular-matrix molecules in the tissues, such as glycosaminoglycans and proteoglycans, may affect the formation and deposition of fibrils.
