 MPTP-induced experimental parkinsonism has been considered as the best model available at present of Parkinson's disease. MPTP is first oxidized to MPDP+ by glial monoamine oxidase B. The MPDP+ undergoes dispropornation reaction to MPTP and MPP+. MPP+ is actively taken up into dopaminergic neurons through an energy dependent reuptake system. Thus the selective toxicity against dopaminergic neurons has been fairly well elucidated. However, the mechanism of neuronal degeneration is not known yet.

 In this communication, proposed hypotheses on mechanism of neuronal degeneration are reviewed.
