MPTPの代謝と毒性発現機構

The mechanism of MPTP neurotoxicity in relation to its metabolism and disposition.

はじめに

　南カリフォルニアで起きたパーキンソン病多発事件をきっかけに，MPTPがヒトに典型的パーキンソン病を起こすことが明らかにされたのは，約4年前の1983年のことである。Langstonら^1）によってなされたこの発見に続き，Burnsら^2）は，MPTPによるパーキンソン病はサルにも再現しうることを示し，MPTPが黒質ドパミン神経を選択的に破壊することを明らかにした。

　これら二つの画期的な研究成果は，パーキンソン病の研究者はもちろんのこと，薬理学者，毒性学者など多くの研究者達の興味をかき立て，MPTPに対する熱狂的研究熱，いわゆる"MPTP fever"をこれらの研究者の間に巻き起こした。その結果，当初はまったく不明であった黒質ドパミン神経に対するMPTPの選択的毒性機構は，ここ数年の内にかなり多くの部分が解明されるに至っている。

MPTP (N-methyl-4-phenyl-1, 2, 3, 6-tetrahydro-pyridine) is a highly selective neurotoxin which appears to degenerate dopaminergic neurons in the nigrostriatal system. Recent accumulation of several years of the studies on MPTP has led us to understand much of the mechanism concerning the selective neurotoxicity of MPTP. MPTP is metabolized by brain monoamine oxidase type B to MPDP⁺, which is further oxidized to the potent neurotoxin MPP⁺. MPP⁺ is selectively taken up by dopamine reuptake system and con-centrated in the nerve cells. The uptake of MPP+ following the metabolic conversion from MPTP appears to account for the selective neurotoxic effects of MPTP on the nigrostriatal dopaminergic neurons.
