MPTP (N-methyl-4-phenyl-1, 2, 3, 6-tetrahydro-pyridine) is a highly selective neurotoxin which appears to degenerate dopaminergic neurons in the nigrostriatal system. Recent accumulation of several years of the studies on MPTP has led us to understand much of the mechanism concerning the selective neurotoxicity of MPTP. MPTP is metabolized by brain monoamine oxidase type B to MPDP+, which is further oxidized to the potent neurotoxin MPP+. MPP+ is selectively taken up by dopamine reuptake system and con-centrated in the nerve cells. The uptake of MPP+ following the metabolic conversion from MPTP appears to account for the selective neurotoxic effects of MPTP on the nigrostriatal dopaminergic neurons.
