 We studied on Creutzfeldt-Jakob disease (CJD) agent in tissue culture cells derived from CJD infected rat brain, in ammonium sulfate fractions, and in chromatograhic fractions. The assay of CJD agent was based on incubation period of intracerebrally inoculated mice. In tissue culture, the cells were fibroblastic and did not express S 100 and glial fibrillary acidic protein. The activity of CJD agent was higher in the cells than in the medium. CJD agent was mostly precipitated with 60% ammonium sulfate solution, although it was still present in 100% ammonium sulfate solution. Sepharose CL-6B gel chromatography revealed that CJD agent was highest in the void and 36000 molecular weight fractions, and were higher in 24000 to 27000 M. W. fraction than in 30000 to 32000 M. W. fraction. CJD agent adsorbed to DEAE cellulose was not eluted with O.1M NaC1, Tris buffer but was fully eluted with 0.3 M NaCl solution. CJD agent should be closely associated with an acidic protein of 36000 molecular weight.
