神経疾患における統計的・集団遺伝学的方法

Statistico-genetic approach in neurological diseases

I．はしがき

　神経疾患にかぎらず問題とする疾患の遺伝性を証明するには，「その家族集積性が遺伝法則と整合性があるか」を検定することが必要である。遺伝性疾患が健康状態と比較して，1遺伝子の存在を仮定することにより，その遺伝様式が説明できる場合を単因子遺伝とかメンデル性遺伝という。単因子遺伝病はヒトの遺伝病のおよそ1割にしかすぎず，残りの大部分は不規則性遺伝病あるいは多因子遺伝病といわれている。単因子遺伝病は直接その家族集積性を調査することで，その遺伝性を検定することが可能である。一方，不規則性遺伝病では，家族集積性だけでなく，遺伝標識として，染色体，HLA多型，DNAの制限酵素切断片多型などを用いて，病気の発症に主効果を顕わす遺伝子の検索も合わせて行なうことで，その遺伝性を確かめることができる場合がある。この論文は紙数に限りがあるので，単因子遺伝病の統計分析の方法を述べ，その実際への利用例として，筆者が行なった日本におけるデュシャンヌ型筋ジストロフィー症の研究を紹介したい。

Abstract

An expository presentation of classical segrega-tion analysis has been made with a special refer-ence to Duchenne muscular dystrophy (DMD). Interpreting that familial aggregations of many neurological diseases would be monofactorial, major genes which might have casused the disease could be identified. By applying segregation analysis to a set of data of DMD in Japan, many difficulties in field survey were examined. It is contended that careful analysis based on genetic model will be helpful greatly to clinicians.
