14-3-2 protein and enolase activity

はじめに

　赤血球中のヘモグロビン，筋肉中のミオシンなどはそれぞれの器官に特異的に存在し，その器官の機能をささえ，特徴づけている蛋白質である。これと同じように神経組織には，その高次な機能をささえる特異蛋白質があるに違いないという観点から，蛋白質分画法の進歩に伴ない，多くの神経組織特異蛋白が発見された^4）。

　1965年Moore ＆ McGregor^38）はウシ脳の可溶性分画をDEAE-セルローズカラムクロマトグラフィーで15分画し，それぞれの分画をさらにstarch gel electrophoresisにて分画して，脳可溶性蛋白の"protein map"を作製した。そしてこのmapと他組織（肝臓）抽出液の"protein map"を比較すると，DEAE-セルローズカラムから高塩濃度下で溶出してくる分画（14，15分画）中に，脳に特異的に検出される蛋白質があることを見出した。14-3-2蛋白はこのうちの一つで，ウシのみならず，広く他種動物の神経組織中にも類似の酸性蛋白が存在し（nervous system specific），それぞれが免疫化学的に共通の抗原性を有する（species non-specific）ことが明らかになった^37）。

　Since the discovery of 14-3-2 protein as a nervous system specific protein, this acidic protein has been the subject of much investigation. Recently, the protein has been shown to be homologous to a subunit of brain specific forms of glycolytic enzyme, enolase. In this report, isolation, characterization and sensitive immunoassay of rat brain enolase isozymes are presented. The immunocytochemical localization and the profiles of enolase isozymes in differentiating cells are also discussed with a special emphasis on the neurobiological significance of the brain specific forms of enolase isozymes.
