GDNFファミリーは四つのメンバーから構成される比較的新しい神経栄養因子群である。GDNFファミリーのシグナルは，それぞれのメンバーに高い親和性で結合する受容体GFRαと，受容体型チロシンキナーゼRETの複合体により伝達される。これらの分子群は，生体内での発現において時間的・空間的に精妙な調節のもとに，特定の神経発生・分化にきわめて重要な役割を果たす。GDNFファミリーの生理機能の解明は，多様な神経栄養因子群の相互作用の理解に不可欠である。

Glial cell line derived neurotrophic factor (GDNF) family of ligands (GFLs) exerts a trophic influence on a variety of neural populations. Although the GFLs are distant members of the TGF-β superfamily, they utilize a unique receptor system in which the receptor tyrosine kinase, RET, and a glycosylphosphatidylinositol (GPI) -lined cell surface protein, GFRa, associate to form a functional receptor. Biochemical attributes in ligand-receptor interaction have been extensively characterized, revealing that there is a stringent pairing between GFLs and GFR αs.
