HGFの神経系における機能と神経疾患治療への展望

Functional roles of HGF in the nervous system: potential application for neural diseases.

肝細胞増殖因子（HGF）は，はじめ肝細胞増殖の本体として見出されたが，実際は多くの細胞に多彩な機能を持つ多機能性因子である。HGFはin vitroで難治性神経疾患に重要な多くの神経細胞（大脳皮質，海馬，中脳ドーパミン作動性や運動ニューロン）や感覚ニューロン，交感神経芽細胞，小脳顆粒細胞などに対し強力な生存，神経突起伸長や遊走促進活性を示す新しい神経栄養因子である。HGFはin vivoにおいて発生過程で交感，感覚神経系や運動神経系で必須の機能を果たすと共に，脳虚血をはじめとする一部の神経疾患モデルですでに著しい治療効果を示すことが明らかになっている。本稿ではとくに神経疾患治療への展望に焦点を当て概説した。

While hepatocyte growth factor (HGF) was initially identified as a potent mitogen for mature hepatocytesresponsible for vigorous regeneration of the liver, it turned to be a well characterized multipotent factor whichtargets a wide variety of cells. Recent extensive studies on expression and functional analysis including knock-out/in mice strategies revealed HGF as a new neurotrophic factor essential for sensory, sympathetic and motorsystem development as well as neural regeneration, such as transient brain ischemia in rat.
