ノックアウトマウス解析の問題点―遺伝的背景をどう揃えるか

Concerning on genetic background of knockout mice for the behavioral studies

ジーンターゲッティング法によって，多くのノックアウトマウスが誕生した。このマウスを使って，行動の解析が行われ，ノックアウトされた分子の高次機能への関与が明らかになりつつある。しかし，問題もある。第一は，脳で働く遺伝子を欠損させると，形態形成が異常となり，脳の機能異常が，形態の異常に隠れて測定できない場合である。第二は，機能的に重複する遺伝子が多く，一つだけの遺伝子ノックアウトでは表現型がでない場合である。ここで取り上げる第三の例は，遺伝的背景の問題である。行動解析では，かなり前から指摘されていたことであるが，突然変異遺伝子の機能を測定した結果が，実は，その遺伝子の効果よりも，遺伝的背景のばらつきによっている例が出始めた。ノックアウトマウスの有用性に関わるこの問題の解決方法について考えてみたい。

Mouse targeted mutagenesis has become a powerful tool for the analysis of brain function. Especially, it has been used to investigate the molecular and cellular mechanisms underlying behaviors such as learning and memory. However, there arises the defficulties to compare the data on behavioral analysis among researchers, since the genetic backgrounds of knockout mice they used are very different among them.
