Glucocorticoids are indispensable for not only maintenance of metabolic homeostasis but also for treatment of a wide variety of human disorders including inflammatory diseases. Glucocorticoids, via binding to the glucocorticoid receptor (GR) in target tissues, exert hormone action. The GR is a ligand-inducible transcription factor, belonging to the nuclear receptor superfamily. Upon binding hormone, the GR dissociates heat shock proteins and translocates into the nucleus, thereby communicating with basal transcriptional machinery, other transcription factors, coactivators and DNA, resulting in the fine tuning of target gene expression.
